摘　　要：目的 :观察铭铂诱导的人卵巢癌细胞A2 780凋亡 ,探讨其抗肿瘤作用机理。方法 :用MTT法测定了铭铂对A2 780的细胞毒作用 ;荧光显微镜从形态学上观察凋亡发生 ,琼脂糖DNA电泳检测程序性细胞死亡特征的DNA降解。结果 :铭铂对A2 780细胞有细胞毒作用 ,半数抑制浓度具有药物接触细胞时间依赖性 ;铭铂处理的A2 780细胞形态学上显示出凋亡细胞特性 ,琼脂糖凝胶电泳显示出特征性程序性细胞死亡DNA梯形图谱。结论 :铭铂能够诱导A2 780细胞凋亡 ,诱导肿瘤细胞凋亡是其一个抗肿瘤作用机理。Objective:To study the anticancer mechanism of minoplatin by observing the induction of apoptosis.Methods:Cytotoxicity of minoplatin was tested by MTT method.Morphological changes of apoptosic cells were investigated by fluorescence microscope.DNA fragmentation was analysed by agarose gel electrophoresis.Results:Minoplatin has distinct cytotoxicity to human ovarian cancer cells,and IC 50 (i e the concentration that reduce the absorbance value to 50% of the control) was related to the time of the drug exposed to cells.Minoplatin-treated ovarian cells showed morphological characteristics of apoptosis.A DNA ladder fragmentation was demonstrated on agarose gel electrophoresis.Conclusion:Minoplatin can induce A2780 cell to apoptosis,and one mechanism of its anticancer effects is to trigger apoptosis.
