原发胃癌中p15基因及蛋白表达的异常

摘　　要：目的检测胃癌组织中p15基因及启动子甲基化状态和P15蛋白表达情况。方法选择p15基因及启动子区域,用PCR-SSCP,MSP(甲基化特异的PCR)法、测序和免疫组化等方法对100例胃癌患者的癌组织和癌旁组织进行检测。结果 11％的病例P15表达阴性,9％的病例具有p15基因启动子区的高甲基化,9％的病例同时有P15表达阴性和p15基因启动子区的高甲基化,p15异常与低分化胃癌有关,p15基因发现两例DNA序列改变。结论 p15基因在胃癌中起一定作用且p15基因启动子区域高甲基化是胃癌中p15基因失活的主要原因。AIM To study the abnormalities of pl5 gene expression and the status of p15 in primary gastric cancers. METHODS The expression of P15 protein were checked with immunochemistry. The deletion/mutation and the methylation of p15 gene were detected by PCR, PCRSSCP, MSP (methylation special PCR) and sequencing. RESULTS Negative staining was found in 11% of the samples, 9% of primary gastric cancer had the hypermethylation of p16, and 9% samples had hypermethylation of p15 and negative expression of P15 protein. In two cases, cancer tissues displayed point mutations in the p15 gene. CONCLUSION The hypermethylation of p15 plays an important role in inactivation of p15 in primary gastric cancer.
