摘  要:目的 探讨甲状腺癌组织中 p5 3基因点突变。 方法 应用非同位素PCR -SSCP技术检测 39例不同类型甲状腺癌石蜡包埋组织中 p5 3基因点突变。 结果  10例低分化甲状腺癌中 4例 (4 0 % )、9例未分化甲状腺癌中 6例 (67% )出现异常电泳 ,表明这些病例相应DNA片段中发生了点突变 ,而 5例乳头状癌和 5例滤泡型癌均未见该基因的突变。 10例出现异常电泳的甲状腺癌中 9例 (90 % )同时呈现 p5 3蛋白的表达。 结论 非同位素PCR -SSCP是一简便、快速、有效的检测基因点突变的方法 ,p5 3基因点突变率与蛋白表达率基本相一致 ,p5 3基因的突变是分化型癌向间变型癌转化的关键性遗传改变。Objective To investigate the point mutation of p53 gene in thyroid carcinoma tissues. Methods Non-isotopic PCR-SSCP method was used to detect point mutation of p53 gene in 29 of thyroid carcinoma.Results The abnormal shifting of the single-strand DNA (ssDNA) was identified in 10 out of 19 cases in anaplastic carcinoma of thyroid gland. Among 10 cases, 4 existed in poor differentiated thyroid carcinomas, 9 in undifferentiated thyroid carcinomas. The point mutation of 10 cases were all found in exons 5-7. 5 papillary carcinomas and 5 follicular carcinomas didn't show point mutation in SSCP analysis.Conclusion Our results indicate that detection of point mutations of p53 gene by PCR-SSCP method is more accurate and reliable than that by immunohistochemistry. The rate of p53 mutation and expression are essential coinsidence. p53 gene mutations is one of the crucial genetic changes triggering the transformation of differentiated into undifferentiated carcinomas in the human thyroid gland.
