ER阴性乳癌细胞株凋亡过程Cyclin DI表达的研究

摘　　要：目的：应用ＤＮＡ末端标记法探讨激素受体阴性乳癌细胞凋亡和Ｃｙｃｌｉｎ　ＤＩ之间的关系。方法：通过１，２５（ＯＨ）２Ｄ３诱发雌激素受体阴性乳癌细胞株ＭＤＡ－ＭＢ－４３５Ｓ凋亡，采用ＭＴＴ法检测细胞增殖情况，ＴＵＮＥＬ法检测细胞凋亡数、免疫组化法检测Ｃｙｃｌｉｎ　ＤＩ，进行相关性分析。结果：在１０－８Ｍ１，２５（ＯＨ）２Ｄ３诱发ＭＤＡ－ＭＢ－４３５Ｓ调亡过程中，癌细胞凋亡率与Ｃｙｃｌｉｎ　ＤＩ表达呈负相关。结论：Ｃｙｃｌｉｎ　ＤＩ的检测可以直接反应雌激素受体阴性乳癌细胞株的凋亡，为临床评价雌激素受体阴性乳癌的治疗效果提供新的指标。Objective：To test the relationship between the expression of Cyclin Dl and apoptosis in the ER (-) breast cancer cell line .Methods：Apoptosis was induced by1,25(OH)2D3in the ER(-)breast cell line.We compared the cell number by MTT,apoptotic cells by TUNEL,and the expression of Cyclin Dl by immunohistochemistry.Results：The expression of Cyclin DI was downregulated during apoptosis in MDA-MB-435s Cell line.It is a negative correlation between the apoptosis and the expression of Cyclin DI .Conclusion ：Cylin DI reflected the effect of treatme nt in ER(-)breast cancer,and it may be a new marker to predict prognosis.
