摘　　要：研究黑暗链霉菌的筛选 ,揭示其退化的主要表征是出现淡黄色菌落 ,退化与时间和菌落表征的关系。比较代表四类不同诱变因子 NTG、Na NO2 、5 - Bu和紫外线的处理效果。认为复壮以紫外线温和诱变最有效。建立了优质孢子富集法。采用原生质体融合——优质孢子富集法——琼脂块法——固体微型发酵法的复合理性筛选。经摇瓶验证 ,获得了比出发菌株高出 2 .5倍的高产菌株。主要成分妥布霉素从 5 2 %上升到71% ,效果显著 。The culture characteristics of Streptomuyces tenebrarius which closely related to the culture degeneraton including changing to a light yellow of colony color, preservation time and the relationship between them were studied. To isolate high tobramycin producing strains, multiple mutagens such as NTG, NaNO 2 and 5 Bu (5 bromouracil), as well as the combined action of UV treatment were used. It reveales that the moderate UV radiation treatment was the best way to culture rejuvenation. In order to further increase the potency of the strain, a measure of enrichment of high quality spores called complex rational screening method which included steps such as protopast fusion, agar block and solid state micro fermentation was used. A new strain which has 2.5 times higher potency than the parent strain with increasing tobramycin component from 52% to 71% was isolated in flasks.
