摘 要:以吸附在硅藻土上的假丝酵母 (Candidasp .16 19)脂肪酶为催化剂 ,在无溶剂体系中 ,研究了月桂酸与海藻糖的酯化反应 .结果表明 ,反应温度为 47℃ ,pH为 7 0 ,月桂酸与海藻糖的摩尔比大于 6时酯化反应的转化率较高 .在 3 2mmol(0 6 4g)月桂酸、0 2mmol(0 0 76g ,即 5 3μl浓度为 1 43g/ml的糖溶液 )海藻糖和 2 0 0mg固定化脂肪酶 (10 0 0U)组成的反应体系中 ,于 47℃振荡反应 36h ,以海藻糖月桂酸双酯计算 ,酯化程度达 95 %以上 .用有机溶剂提取的反应产物经薄层色谱提纯后进行13CNMR分析 ,结果表明 ,产物主要是 6 ,6’ 海藻糖月桂酸酯 ,其纯度为 90 %~ 95 %Reaction conditions for the esterification in a solvent-free system using lauric acid as the acyl donor and trehalose as the acyl acceptor, immobilized lipase from Candida sp. 1619 as the biocatalyst were investigated. The optimal temperature and pH for the esterification were 47degreesC and 7.0, respectively. The conversion rate was enhanced greatly when the molar ratio of lauric acid to trehalose was higher than 6. The reaction was carried out in a 20 ml open conical flask shaken at 47degreesC with 3.2 mmol (0.64 g) lauric acid, 0.2 mmol (0.076 g, 53 mul of 1.43 g/ml trehalose solution) trehalose, and 200 mg immobilized lipase (1000 U). After reaction for 36 h, about 95% of trehalose was esterificated based on the diester of trehaloselaurate. The products were purified by thin layer chromatography and identified by C-13 NMR as 6,6'-trehaloselaurate with a purity of 90% similar to 95%.
