摘 要:目的 探讨肺癌组织中p5 3基因蛋白表达和肺癌患者血白细胞p5 3基因突变及其相互关系。方法 应用免疫组织化学S -P法检测肺癌组织中p5 3基因蛋白表达及聚合酶链反应 -单链构象多态性分析 (PCR -SSCP)检测其中肺癌患者血白细胞p5 3基因突变。结果 p5 3基因蛋白在 5 1例肺癌组织中的阳性表达率为 5 9% ,其中p5 3蛋白在鳞癌、腺鳞癌和腺癌的表达差异无显著性 (P >0 .0 5 ) ,但有淋巴结转移者 ( 77% )显著高于无淋巴结转移者 ( 4 5 % ,P <0 0 5 ) ,p5 3蛋白在肺癌癌旁正常肺组织及白细胞中均为阴性 ;5 1例肺癌患者中 2 8例血白细胞DNA均未见p5 3基因突变。结论 p5 3基因在肺癌患者血白细胞DNA未见突变 ,p5Objective To study the relationship between p53 protein expression in lung cancer tissues and p53 gene mutation in white blood cells in lung cancer patients. Methods Immunohistochemistry (S-P) was used to detect p53 protein expression in cancer tissue and PCR-SSCP for p53 gene mutation in white blood cells of lung cancer. Results The positive rate of p53 protein was 59% in lung cancer tissue. There was no significant difference in p53 expression among squamous carcinoma, adeno-squamous carcinoma and adenocarcinoma( P >0.05), it was significantly higher in patients with lymph node matastasis (77%) than without ones (45%, P <0 05),p53 protein expression was negative in normal lung tissue ajacent to cancer focus. There were no any p53 mutations in white blood cells DNA of 28 patients. Conclusion There are no any p53 mutation in white blood cells DNA of patients with lung cancer, the lung cancer patients with positive expression of p53 may predispose to have lymph node matastasis.
