大肠癌组织P21^(WAF1/CIP1)的表达与p53基因突变的关系

摘　　要：目的 :探讨大肠癌组织P2 1WAF1/CIP1的表达与p5 3基因突变的关系及其意义。方法 :应用RT -PCR、RCR -SSCP方法对 42例新鲜大肠癌组织中上述两种基因的表达进行研究。结果 :p5 3基因在大肠癌组织中的突变率为47.6 % (2 0 / 42 ) ,且在中、低分化大肠癌组织中的突变北显著高于高分化者 (P <0 .0 5 ) ;P2 1WAF1/CIP1在中、低分化大肠癌组织中的表达显著低于高分化癌组织 (P <0 .0 1)。 2 0例p5 3基因突变组中 ,P2 1WAF1/CIP1表达阳性率明显低于 2 2例p5 3基因未突变组 (P <0 .0 5 )。结论 :大肠癌组织中P2 1WAF1/CIP1的低表达可能与p5 3基因突变有关。Objective To investigate the relationship between the expression of P21 WAF1/CIP1 and p53 genomic mutation in colorectal carcinoma. Methods RT_PCR, PCR_SSCP was used to study two_gene expressions above of 42 cases of colorectal carcinoma fresh tissue.Results The mutation rate of p53 in carcinoma tissue was 47.6%(20/42), and mutation rate in poorly or moderately differentiated colorectal carcinoma was significantly higher than that of highly differentiated P <0.05, but the expression of P21 WAF1/CIP1 in the former was lower than the latter P <0.01, and the expression of P21 WAF1/CIP1 in group of 20 case of p53 mutation was lower than that in group of 22 case of p53 non-mutation( P <0.05) Conclusion Low expression of P21 WAF1/CIP1 in colorectal carcinoma was probably related to the mutation of p53 gene.
