摘  要:目的 观察蝎毒抗癌多肽(APBMV)对人肝癌细胞SMMC-7721的作用。方法MTT法、生长抑制实验和集落形成实验。结果 APBMV对 SMMC-7721细胞具较强毒性作用,IC50为11.33μg/ml;APBMV可显著抑制细胞生长,剂量与时间效应关系明显。结论APBMV是东亚钳蝎蝎毒中有效低毒的抗肿瘤成分,对肝癌细胞有一定的毒性作用。Objective To obserne the effect of antineoplastic polypeptide from Buthus martensii venom (APBMV) on the human heptoma cells SMMC-7721. Methods Using the MTT colorimetric method, the growth-inhibiting test and the colony-formation inhibiting assay. Results The results revealed that APBMV had obvious cytotoxicity on SMMC-7721,1C50 was 11. 33μg/ml;APBMV could obviously inhibit the cellu- lar growth. There were obvious dose-response and timeresponse relationship. The IC50 were 15. 8T54ig/ml, 13. 05μg/ml,8. 70μg/ml,respectively at 24h,48h and 96h intervals after SMMC-7721 cells were exposed to APBMV. There were instinct correlations in inhibiting effect between24h and 48h, or 48h and 96h(rrr9709,P<0. 05;r0. 9831,P<0. 02). When the concentration of APBMV exceeded 8μg/ml,the colony- formation numbers of cells reduced dramatically(P<0. 01) ,1C50 was 10. 83μg/ml. Conclusion APBMV was effective and low toxicity antineoplastic component in the venom of Buthus Martensii scorpiton. It posses certain toxicity on hepatoma cells.
