动态浊度法定量检测大蒜注射液中细菌内毒素

摘　　要：目的 :分析研究大蒜注射液对鲎试剂的干扰作用 ,探讨以细菌内毒素检查法取代热原检查法检测大蒜注射液的可行性。方法 :本试验采用检测细菌内毒素的动态浊度法 ,用EDS 99细菌内毒素测定仪对大蒜注射液进行了抑制增强试验。结果 :样品经 1∶2 0倍稀释 ,平均回收率在 5 0 %～ 2 0 0 %范围内 ,标准曲线的回归方程为logT =2 .82 47- 0 .32 5 2 5logC ,r =- 0 .99875 ,表明样品溶液对TAL试剂反应无干扰作用。结论 :凝胶法检测大蒜注射液 ,干扰作用较大。采用动态浊度法进行定量检测 ,干扰因素小 ,可以作为日常检测使用。OBJECTIVE To approach the feasibility of replacing pyrogen test with bacterial endotoxin test (BET) by analysing the interference of the injection to Limulus Amebocyte Lysate (LAL). METHODS Kinetic turbimetric assay (KTA) is used to determinate the bacterial endotoxin and have an inhibition and strengthen test by using of the EDS 99 bacterial endoxin systems.RESULTS It is diluted to 1:20.The regression equation shows that the sample solution has no interference to LAL.CONCLUSIONS KTA can be used with less interference.
