摘  要:为了解市售猪肉中沙门氏菌耐药基因和毒力基因携带状况,以及PFGE分子分型情况,用PCR方法对分离的22株沙门氏菌携带的耐药基因和毒力基因进行检测,并运用XbaⅠ酶进行酶切后完成PFGE分析,再利用BioNumerics软件对分离菌株的指纹图谱进行聚类分析。结果显示:分离菌株对氨苄西林的耐药性最严重,耐药率为72.73%,其次是对氟苯尼考,耐药率为68.18%;在10种耐药基因检测中,共检出9种;除喹诺酮类药物外,分离菌株耐药基因与耐药表型的符合率为95%,其中88%的分离菌株携带酰胺醇类耐药基因(catI)和β-内酰胺类耐药基因(blatem-1/blapsE-1/blaCMY-1/blaoxA-1)。在14种毒力基因检测中,共检出13种;所有分离菌株均携带mgt C和bcfA毒力基因,其次是mogA、araB、stn、fimA毒力基因,携带率均为95.45%;22株沙门氏菌共分为10个群,包括8个带型,菌株相似度为60%~100%。结果表明:市售猪肉中的沙门氏菌携带多种耐药基因和毒力基因,须采取措施防止沙门氏菌污染猪肉产品。In order to recognize the carrying status of drug-resistance genes and virulence genes of Salmonella in retail pork products, and to study the PFGE typing situation, the resistance and virulence genes carried by 22 isolated Salmonella strains were detected by PCR and digested with XbaI enzyme. Then the PFGE analysis was completed, and the fingerprints Of the isolated strains were clustered by BioNumerics software. Results showed that these isolates had the most serious resistance to Ampicillin, the drug resistance rate was 72.73%, followed by Florfenicol, the resistance rate was 68.18%. In the detection of 10 kinds of drug resistance genes, a total of 9 were detected. Except for Quinolones, the coincidence rate of drug-resistant genes and drug-resistant phenotypes was 95%, and 88% of the isolates carried Amphenicol-resistant genes (cat1) and 13 -lactam-resistant genes (blatem-1/blapsE-1/blaCMY-1/ blaoxA-1 ) . Among the detection of 14 kinds of virulence genes, 13 of them were detected. The carrying rates of virulence genes ofmgtC and bcfA were the highest (100%) , followed by mogA, araB, stn andfimA gene, and the carrying rates were all 95.45%. These isolates could be divided into I0 groups, including 8 different bands, and the similarity was at the range of 60% to100%. The results indicated that, the Salmonella in the retail pork carried a variety of drug-resistance and virulence genes, which posed a serious threat to the health of consumers. Hence, measures should be taken to prevent Salmonella contamination, so that meat safety could be ensured.
