摘  要:益生菌是通过多种作用机制来提供功效作用,其中包括提供胃肠道保护屏障,改变肠道内环境p H来提供非致病菌的增殖,从而增强宿主的免疫应答能力,产生抗菌物质与其在受体肠道中进行竞争。乳酸菌对于宿主肠道上皮细胞的黏附性具有菌株特异性而并非宿主特异性,由此可以采用体外模型来筛选所诉求的益生菌。双歧杆菌的黏附过程首先是通过非特异性接触,之后与受体结合,实验过程主要是通过菌株对胞外机制的黏附能力进行筛选具有高黏附特性的Bifidobacterium lactis。采用20株分离自健康人体肠道的Bifidobacterium lactis进行筛选,并进一步从具有功能性的菌株进行了高黏附性筛选、耐酸耐胆盐等筛选,其中筛选出1株各方面均具有良好抗逆性的Bifidobacterium lactis BLa80,对肠道上皮细胞的黏附率达到1.1%,胆固醇降解能力达到34.56%;非外源性保护条件下胃酸p H环境下的存活率达到21.47%,胆盐耐受性达到29.3%。所筛选出的菌株经过中国微生物研究所鉴定为乳双歧杆菌Bifidobacterium lactis,并命名为BLa80。Probiotics provide efficacy through a variety of mechanisms of action,including providing a protective barrier to the gastrointestinal tract,altering the pH of the intestinal environment to provide PrOliferation of non-pathogenic bacteria,thereby enhancing the host's immune response,producing antibacterial substances and competition in the intestinal tract.The adhesion of lactic acid bacteria to host intestinal epithelial cells is strain specific and not host specific,and an in vitro model can be used to screen the desired probiotics.The adhesion process of Bifidobacteria firstly passes through non-specific contacts and then binds to the receptor.This experiment mainly screens Bifidobacterium lactis with high adhesion properties by the ability of the strain to adhere to extracellular mechanisms.Twenty strains Of Bifidobacterium lactis isolated from healthy human intestinal tract were screened.Furthermore,high-adhesive screening,acid-tolerant and bile-salt-resistant screening were carried out from functional strains, and a Bifidobacterium lactis with good stress resistance in all aspects was selected,and the adhesion rate of BLa80to intestinal epithelial cells was selected.It reached1.1%,the cholesterol degradation ability reached34.56%;the survival rate under the environment of gastric acid pH under non-exogenous protection conditions reached21.47%,and the tolerance of bile salt reached29.3%.The selected strain was identified as Bifidobacterium lactis by the Chinese Institute of Microbiology and named as BLa80.
