摘 要:目的:研究芦荟蒽醌对高脂血症小鼠脂代谢及脂蛋白酯酶(LPL)、微粒体甘油三酯转运蛋白(MTTP)基因表达的影响。方法:采用灌胃高胆固醇乳剂的方法建立高脂血症的模型,用精制芦荟蒽醌对高脂血症小鼠进行干预,持续4周后测定血清中总胆固醇(TC)、甘油三酯(TG)、高密度脂蛋白胆固醇(HDL-C)和低密度脂蛋白胆固醇(LDL-C)等生化指标,以此评定芦荟蒽醌对高脂血症小鼠的降血脂作用;采用实时荧光定量RT-PCR法测定小鼠心脏、肝脏、脾脏、肺脏和肾脏等组织中LPL和MTTP mRNA的表达水平,从分子水平探讨芦荟蒽醌的降血脂机制。结果:芦荟蒽醌具有良好的降血脂作用,能够显著降低高脂血症小鼠血清中的TC、TG和LDL-C水平(P<0.01),显著升高HDL-C水平(P<0.05或P<0.01);芦荟蒽醌可以显著提高心脏、肺脏及肾脏中LPL基因m RNA表达量(P<0.05或P<0.01),显著降低肝脏中MTTP基因mRNA表达量(P<0.01)。结论:芦荟蒽醌具有调节高脂血症小鼠LPL和MTTP基因表达的作用,进而起到缓解高脂血症的功效。Objective: The effects of aloe anthraquinones on lipid metabolism and expression of lipoproteinlipase(LPL) and microsomal triglyceride transfer protein(MTTP) genes in hyperlipidemia mice were studied in this experiment.Methods: The model of hyperlipidemia was caused by intragastric administration of high cholesterol emulsion. The hyperlipidemia mice were intervened with refined aloe anthraquinones. After the refined aloe anthraquinones were fed to mice four weeks, the biochemical indicator such as total cholesterol(TC), triglyceride(TG), high-density lipoprotein cholesterol(HDL-C) and low-density lipoprotein cholesterol(LDL-C) in serum were detected in order to evaluate the effect of aloe anthraquinone on reducing blood lipid in hyperlipidemia mice. The LPL and MTTP m RNA in the mice tissues of heart, liver, spleen, lung and kidney were measured by real time RT-PCR after the lipid-lowering animal experiment.The mechanism of reducing blood lipid by aloe anthraquinone was discussed from the molecular level. Results: The results showed that aloe anthraquinone has good hypolipidemic effect. It can significantly decrease the TC, TG and LDL-C levels(P<0.01) and significantly increased the HDL-C levels(P<0.05 or P<0.01) in serum of hyperlipemia mice. Aloe anthraquinone can significantly increase LPL gene mRNA expression in heart, lung and kidney(P<0.05 or P<0.01) and can significantly decrease MTTP gene mRNA expression in liver(P<0.01). Conclusion: These results suggest that aloe anthraquinone can regulate the expression of LPL and MTTP gene in hyperlipidemia mice, and then contribute the effect of relieving hyperlipidemia.
