摘 要:目的:研究菊粉复配灵芝多糖对HepG2细胞胰岛素抵抗的调节作用。方法:采用高糖高脂培养HepG2细胞,建立胰岛素抵抗模型(IR-HepG2细胞模型)后,用菊粉、灵芝多糖及其复配物继续培养细胞24h,测定细胞葡萄糖消耗量、糖原合成量、细胞内超氧化物歧化酶(superoxide dismutase,SOD)和谷胱甘肽过氧化物酶(Glutathione peroxidase,GSH-Px)活力。结果:与模型组相比,菊粉和灵芝多糖均可显著提高IR-HepG2细胞的葡萄糖消耗量、糖原合成量、SOD和GSH-Px活力,并呈现剂量依赖性。将菊粉和不同浓度的灵芝多糖复配之后,与单一的菊粉组相比,复配高剂量组的葡萄糖消耗量、糖原含量、SOD和GSH-Px活力均极显著升高(P <0.01)。结论:菊粉复配灵芝多糖可显著促进IR-HepG2细胞葡萄糖的利用,增加糖原合成,提高SOD和GSH-Px酶活力,改善糖代谢紊乱。Objective :To investigate the effect of inulin combined with ganoderma lucidum polysaccharides on insulin-resistance of HepG2 cells. Methods :The insulin-resistant models of HepG2 cells(IR-HepG2)were induced by high concentration of glucose and palmitic acid for 36 hours. Then,the inulin and ganoderma lucidum polysaccharides were added into the cultured medium. After 24 h hours,the glucose consumption,the glycogen synthesis,superoxide dismutase(SOD)and Glutathione peroxidase(GSH-Px)activities of each group cells were measured. Results :Compared with the model group,the glucose consumption,the glycogen synthesis,SOD and GSH-Px activities of inulin groups and ganoderma lucidum polysaccharides groups increased significantly(P< 0.05). Compared with the single inulin group,the glucose consumption,the glycogen synthesis,SOD and GSH-Px activities of combined groups increased significantly(P < 0.01). Conclusion :The inulin combined with ganoderma lucidum polysaccharides can improve the glucose consumptions,increase glycogen synthesis,and enhance SOD and GSH-Px activities in insulin-resistance model of HepG2 cells.
