摘  要:为了解2017年冬季上海临床接诊犬的流感病毒感染情况和病毒变异趋势,收集上海某动物医院疑似犬流感样品进行RT-PCR检测,构建HA、NA的重组载体并测序,对获得的序列进行进化分析、氨基酸变异分析,综合我国同亚型犬流感病毒HA序列进行进化率分析。结果显示:23份疑似样品中检出5份阳性样品,获得5个H3亚型的HA序列、2个N2亚型的NA序列,上述序列在进化树中已经形成独立的进化分支,与韩国、广东毒株亲缘关系最近;氨基酸比对表明,HA和NA在重要位点发生许多一致的突变,如抗原位点区域、糖基化位点等,NA茎部未见氨基酸插入;对2007年至2017年中国南方地区分离的H3N2犬流感病毒HA片段进行进化率、变异率分析显示,年平均进化率呈逐年增高趋势,变异不断积累。研究表明,开展犬流感病毒变异监测,掌握病毒变异和进化方向,对防控该病、防止病毒向人群传播有重要意义。Canine influenza virus( CIV) is a newly emerged infectious disease. However,little is known about the prevalence and variation of this virus in pet dogs,and this study was aimed at the surveillance of CIV infection in a veterinary clinic of Shanghai. Five positive samples were obtained by RT-PCR from 23 nasopharyngeal swabs. Five HA segments of H3 subtype and 2 NA segments of N2 subtype were sequenced using a recombinant T-vector. From the phylogenetic trees,the H3 segments identified in this study were most similar to H3N2 viruses isolated in Korea and Guangdong Province and were clustered into an independent clade,and so were the N2 segments. Comparison of the deduced amino acid sequences showed that all the segments had several mutations at important positions. In HA1,there were some substitutions at the antigenic sites,glycosylation sites and cleavage sites. As for NA,no insertion in stalk was found. Furthermore,calculation with HA sequences isolated in southern China from 2007 to 2017 revealed that the average evolution rate increased in the line chart indicating accumulation of antigen drift. Overall,these data indicated that in-depth study was required for a better understanding of the variation of CIV and control of its potential threat to public health.
