应用阳离子脂质体及裸HSV-TK基因瘤体内直接注射比较研究

摘　　要：目的　比较研究阳离子脂质体及裸HSV TK基因瘤体内直接注射治疗肝癌的效果 ,探讨较好的基因转移方法。方法　用阳离子脂质体Lipofectin tk基因复合物及裸pLXT于小鼠瘤体内直接注射 ,观察肿瘤的生长情况。结果　瘤体内基因转染后脂质体介导的pLXT、ACV治疗组及裸pLXT注射、ACV治疗组平均瘤体积均显著小于其它六组对照组 (P <0 0 0 1,P <0 0 5 ) ,前两者比较亦有显著差异 (P <0 0 2 )。结论　阳离子脂质体介导的HSV tk基因转移的方法简便、安全、有效。裸tk基因瘤体内直接注射效果亦很明显。Aim To compare the effect of antiheptocellular carcinoma by in vivo direct intratumal injection of lipofectin tk and nude HSV tk gene.Methods Lipofectin tk and nude HSV tk gene were directly injected into murine H 22 hepatoma,and the size and weight of tumor was observed.Results There was a significant reduction of tumor size as compared with that in other six control groups ( P<0 001,P<0 05 ).Between animals treated with liptk ACK and tk ACV,there was a significant difference ( P <0 02)).No significant difference was observed in terms of growth rate of tumors among other six control groups ( P <0 05).Conclusion The approaches of in vivo direct injection of cationic liposome tk and nude HSV tk are all simple,safe and effective,and the former is much better than the latter.
