摘  要:为了探讨糖皮质激素的抗痫效应 ,分别用成年大鼠以及培养的新生大鼠的神经细胞 ,在经谷氨酸钠和地塞米松处理后 ,进行免疫细胞化学染色。观察其代谢性谷氨酸受体 1(m GL U R1)免疫反应性的变化 ,并对实验结果进行图像分析。结果发现 :经谷氨酸钠致痫处理后 ,大鼠海马 CA1区以及培养的神经细胞的 m GL UR1免疫反应阳性细胞均较正常对照组增多 ,平均光密度值增加 ;再经地塞米松处理后 ,m GL UR1免疫反应阳性细胞则较致痫组减少 ,平均光密度值减少。上述改变经统计学分析均有显著性差异。结果提示 :地塞米松可以抑制谷氨酸钠诱发的癫痫发作 ,其作用途径之一可能是通过抑制 m GL UIn order to investigate the anti-epileptic effect of glucocorticoid, the changes of metabotropic glutamate receptor 1 (GLUR1) immunoreactivity were observed in epileptic rats and cultured nerve cells after treated with glutamate and dexamethasone. The immunocytochemistry method was used in this experiment. By the image analysis, the results showed that mGLUR1 positive cells and the average optical density were increased in both adult rats and cultured nerve cells after the treatment of glutamate; while after the treatment of dexamethasone following glutamate, they were both decreased. The difference was significant statistically. The results suggested that dexamethasone could inhibit the glutamate-induced epileptiform and its anti-epileptic effect might be relevant to the inhibition of the mGLUR1 expression.
