摘  要:优化了嗜碱芽孢杆菌(Bacillus alkalophilus)木聚糖酶、里氏木霉(Trichoderma reesei)木聚糖酶和Pulpzyme HC 2500木聚糖酶3种木聚糖酶对麦草碱性亚硫酸盐制浆(ASP)废液的酶解工艺,并比较了它们的酶解特性。结果表明:嗜碱芽孢杆菌木聚糖酶在酶用量6.0μmol/(mL·min)、pH7.0、温度50℃、时间8 h和废液质量分数50%的条件下,相对多糖水解率为27.7%;里氏木霉木聚糖酶在酶用量10.0μmol/(mL·min)、pH5.0、温度50℃、时间8 h和废液质量分数50%的条件下,相对多糖水解率为48.5%;Pulpzyme HC 2500木聚糖酶在酶用量8.33μmol/(mL·min)、pH8.0、温度55℃、时间4 h和废液质量分数50%的条件下,相对多糖水解率为48.0%。比较认为,Pulpzyme HC 2500木聚糖酶具有较高的酶解效率,经该酶酶解处理,ASP废液中还原糖含量由1.2 g/L提高到11.1 g/L。大分子聚糖的降解有利于后续木质素磺酸盐的分离提取和利用。The wheat straw alkaline sulfite pulping( ASP) is hopeful to be a potential non-wood lignocelluloses biomass biorefinery process. However,ASP spent liquor utilization is a bottleneck towards the biorefinery. In the paper,the enzymolysis conditions and properties of three kinds of xylanases from Bacillus alkalophilus,Trichoderma reacei and Pulpzyme H C were investigated based on the relative polysaccharide hydrolysis rate( RPHR). The results indicated that 27. 7% of the RPHR was obtained by the xylanase hydrolysis of Bacillus alkalophilus at 6. 0 μmol /( mL·min) of enzyme dosage,50% of spent liquor concentration( weight percentage),pH 7. 0,50 ℃ for 8 h. 48. 5% of the RPHR was found by the enzymolysis of Trichoderma reesei at 10. 0 μmol /( mL·min) of enzyme dosage,50% of spent liquor concentration,pH 5. 0,50 ℃ for 8 h. The RPHR of Pulpzyme HC xylanase reached to 48. 0% at 8. 33 μmol /( mL·min) of enzyme dosage,50% of spent liquor concentration,pH8. 0,55 ℃ for 4 h. The reducing sugar content of ASP spent liquor reached to 11. 1 g/L from 1. 2 g/L by the enzymolysis of Pulpzyme HC. Pulpzyme HC was more powerful and popular comparing with Bacillus alkalophilus and Trichoderma reesei in aspects of the efficiency and industrial application conditions. The enzymolysis of polysaccharide in ASP spent liquor was benefit to the following lignosulphonate separation and utlization.
