摘  要:羊栖菜多糖是从马尾藻科植物羊栖菜 (Sargassumfusiforme(Harv .)Setchel)中提取到的 ,具有抗肿瘤、抗凝血、降血糖、提高机体免疫功能等生理活性 .本文研究用水提法提取羊栖菜多糖 ,考虑到提取温度、时间、加水量等因素的影响 ,优化提取条件 ,确定水提法的最佳温度 80℃ ,提取时间 3h ,水加量为羊栖菜样品的 3倍 .并采用乙醇沉淀、透析及凝胶层析等方法对羊栖菜提取物中多糖进行沉淀、提纯和分离获得 4个组分 ,并用光谱扫描法、凝胶色谱和旋光度法鉴定其纯度 .To make best use of the precious marine resources, the methods of extraction and purification polysaccharide from Sargassum fusiformeg was studied in this paper. The conditions of extracting SFP by boiling water was investigated, result shows that the optimum extraction temperature is 80 ℃, optimum time is 3 h. A water soluble rude polysaccharide was purified by further fraction precipitation with ethanol. The abstract was fractionated by anion exchange chromatography(DEAE Sephadex A 25) column and isolated on membrane. The column was eluted with linear gradient of 0.65 mol/L, 1.00 mol/L, 1.35 mol/L sodium chloride and the three major fraction F 1?F 2?F 3 were obtained. After purification, four types of polysaccharide were obtained from Sargassum fusiforme which were f 1?F 1?F 2?F 3. Analyzing the all fraction of SFP, low molecular weight SFP Fraction (f 1) and high molecular weight SFP fraction (F 1) were tested for their biological activities.
