摘  要:目的 了解镉和汞对细胞hprt基因位点的影响 ,探讨化学诱变物致机体损伤早期检测的敏感指标。方法 采用多核细胞法研究化学诱变物氯化汞和镉对人血淋巴细胞hprt基因位点的影响及突变频率。结果 氯化汞可诱发大鼠血淋巴细胞hprt基因位点突变 ,突变频率随着染毒浓度的增加而升高 ,突变频率 ^Y (10 -3 )与氯化汞浓度C (mg/kg)之间拟合成 ^Y =0 0 4 2 3C +1 0 4 6 3的直线回归方程。不同浓度金属镉也能致hprt基因突变 ,镉浓度与突变频率之间有剂量反应关系。 结论 氯化汞和镉对血淋巴细胞hprt基因位点及突变频率有影响 。Objective To study the effect of Cd and HgCl 2 on hprt gene in blood lymphocytes,and explore a sensitive index in chemical mutants on body hazard at early monitoring.Methods A simple,rapid and sensitive poly nuclear cell assay was used to investigate the frequency of hprt locus mutation both in vivo and in vitro by Cd and HgCl 2 and its dose\|effect relationship.Results\ The results showed that HgCl\-2 could induce hprt locus mutation in mice lymphocytes.The frequency of hprt locus mutation incrcased linearly with the doses expressed in a linear model:= 0 042 3C + 1 046 3 .The Cd could induce hprt locus mutation in human lymphocytes.The frequency of hprt locus mutation and the doses showed a dose\|response relationship.Conclusions Cd and HgCl 2 could result in hprt locus mutation in lymphocytes and the frequency of hprt locus mutation increased linearly with the doses.It could be expected that the frequency of hprt locus mutation might be used as a sensitive index at early monitoring in the exposed population.
