摘  要:目的对酶法提取海狗肾多肽的工艺条件进行研究。方法以多肽含量为指标,考察了4种蛋白酶对海狗肾蛋白的酶解能力,选取A.S1398中性蛋白酶为试验用酶,利用单因素试验和正交试验,分别对海狗肾多肽提取工艺中加水量、加酶量、酶解温度、酶解时间进行优化。结果最佳酶解工艺条件为:加水量为9倍,加酶量为1.5%,温度50℃,时间4 h,pH 7.0。结论在此条件下,海狗肾多肽得率为48.75%,工艺重复性好,适合规模化生产。Objective To study the enzymatic extraction process of seal kidney peptide.Methods With seal kidney peptide content as an indicator to investigate the ability of four kinds of protease enzyme,then we found neutral protease enzyme A.S1398 was the best.Through single factor experiments and orthogonal experiments,we respectively tested the amount of water,enzyme dosage,reaction temperature,reaction time of seal kidney peptide extraction process,which was afterwards optimized.Results Enzyme extraction process of seal kidney peptide was determined:9times of water and 1.5% of enzyme quantity to material quantity,50 ℃ of temperature,4 h of reaction time,pH 7.0.Conclusion Under the above conditions,the yield of seal kidney polypeptide is 48.75%.The technique is reproducible,furthermore,could meet large-scale industrial operation needs.
