摘  要:天然产物生物合成途径中的"隐藏"的修饰反应有着新颖的酶催化机制和特殊的生物学意义,文章概述了几种不同类型的隐藏反应方式,包括卤化、甲酯化、氨基酰化、脂肪酰化和磷酸化5类。分析并总结"隐藏"的修饰反应对于我们认识天然产物生物合成途径的复杂性和多样性,推动天然产物生物合成领域的研究具有重要意义。同时也对新酶学机制的发现和利用基因工程手段来改造新药打下良好的基础。Natural products are the most important components in modern drug industry. In recent years, the research on natural product biosynthesis has become more and more deep and about more than three hundred antibiotic biosynthetic gene clusters have been cloned. The cryptic reactions reported in natural product biosynthetic pathway usually have novel enzymology mechanisms and biological significances. Here,this review summarizes five cryptic modification patterns existing in different antibiotics biosynthetic pathways,including halogenation, fatty acylation, methyl esterification, aminoacylation and phosphorylation modifications. The functions and mechanisms of these cryptic modification have also been described, showing six different reaction patterns, in which the two important reactions, the cyclopropane and the N, C-bipyrrole moieties are catalyzed by cryptic halogenation. Analyzing and summarizing the cryptic reactions of natural product biosnthetic pathways could help us understand the structure diversities of natural products, lay the foundation for our study of the mechanism of the novel natural product biosynthesis and pave the way for novel drug discovery in future clinic use.
