摘 要:细胞和各种生物大分子如核酸、蛋白质等的分离和纯化已逐渐成为生命科学领域内必不可少的技术手段。分离纯化技术的发展对促进整个生物学的发展具有举足轻重的作用。磁性微球分离技术是基于固相磁性材料分离特性,综合生物分子、细胞等因素而发展出的一种新型分离技术。该技术能够简便、快速、柔和、高纯度地分离所需物质。近年来,磁性微球已逐步应用于生物医学、细胞学和生物工程等领域。论文主要回顾了国内外近5年的文献,介绍了磁性微球的结构、特性和分离原理,并详细论述了磁性微球在细胞分离和分类、蛋白质提纯、酶固定化、核酸分离以及免疫分析与检测等领域的应用现状,最后总结并展望了磁分离技术在生物分离纯化和检测中的应用前景。The separation and purification of cells and a variety of biological macromolecules such as nucleic acid,protein are indispensable part of the life sciences.The development of separation and purification technology plays a decisive role for the entire biology.Magnetic microsphere separation is a new type of separation technology based on solid-phase magnetic separation characteristics and integrated biological factors,such as biological molecules and cells.The high-purity target products can be isolated and purified by means of this technology conveniently,quickly and softly.In recent years,magnetic microspheres were progressively applied in many fields,such as biomedicine,cytology and separation engineering.In this paper,the nearly five years of literatures at home and abroad were reviewed in the structure,characteristics and separation principle of magnetic microspheres.The present application situation of magnetic microspheres in cell separation and classification,protein purification,immobilized enzyme,DNA separation and immune analysis and detection were detailedly discussed,and finally the application prospects of magnetic separation technology in of biological purification and detection were summarized.
