摘  要:目的对大蒜素诱导卵巢癌耐顺铂细胞凋亡机制进行初步探讨,从而为大蒜素成为临床耐顺铂卵巢癌患者的有效治疗药物提供实验依据。方法首先对卵巢癌耐顺铂细胞株SKOV-3/DDP进行培养,选取对数生长期的SKOV-3/DDP细胞,将12.5μg/ml的大蒜素作用24h、48h、72h后,用流式细胞仪检测各组细胞周期的改变。结果与不加药的对照组比较,12.5μg/ml的大蒜素作用24h、48h、72h后,SKOV-3/DDPG0/G1期细胞减少,G2/M期细胞明显增加,而且呈现时间依赖性,提示SKOV-3/DDP细胞经大蒜素处理后细胞周期阻滞于G2/M期。结论大蒜素将SKOV-3/DDP细胞阻滞在G2/M期,可能是大蒜素诱导SKOV-3/DDP细胞凋亡的机制之一,这为大蒜素可成为临床耐顺铂卵巢癌患者的有效治疗药物提供了实验依据。Objective To carry out a preliminary study of allicin - induced cisplatin - resistant ovarian cancer apoptosis mecha- nism to provide the appropriate reference for the clinical treatment of cisplatin - resistant ovarian cancer patients. Methods First, we cultivated cisplatin - resistant ovarian cancer cell lines SKOV - 3/DDP and then selected SKOV - 3/DDP when it reached logarithmic phase. After its exposure to allicin at the concentration of 12.5 μg/ml for 24h,48h, and 72 h, flow cytometry was used to detect the change of the cell cycle. Results Compared to the control group without dosing,flow cytometry showed that after SKOV -3/DDP′s exposure to allicin at the concentration of 12.51μg/ml for 24h,48h, and 72 h, the amount of cells decreased in G0/G1 phase and increased significantly in the G2/M phase, and cells were time dependable,which suggested allicin caused arrest of SK- OV - 3/DDP cells in G2/M phase.. Conclusion Allicin causing arrest of SKOV - 3/DDP cells in the G2/M phase may be one of the mechanisms of which allicin induces SKOV - 3/DDP cell apoptosis, and this has provided the appropriate reference for allicin to become an effective drug for clinical treatment of cisplatin - resistant ovarian cancer.
