摘 要:目的研究HBsAg阳性肝癌中PTEN基因的表达,分析PTEN蛋白表达与肝癌病理类型、分化程度的关系。方法采集2009年—2011年在滨州医学院附属医院住院治疗的HBsAg阳性的原发性肝癌29例患者血液及术后新鲜肝组织,通过RT-PCR扩增PTEN基因,采用免疫组织化学法检测羊抗人PTEN多克隆抗体。结果 PTEN基因位于254bp处PTEN基因扩增条带及380bp处GAPDH内参基因条带。肝癌组织中PTEN阳性检出率为69.0%(20/29),相对表达量为0.713±0.125。PTEN蛋白免疫组化表达结果显示PTEN阳性染色呈棕黄色颗粒状或片状,均定位于细胞质中。在表达程度上,肝癌组织PTEN蛋白表达(+)为4例,(++)为13例,(+++)为5例。肝细胞癌组与胆管细胞癌组的PTEN蛋白强阳性表达例数分别为16例、2例,两组存在差距(χ2=4.398,P<0.05),即病理类型与PTEN蛋白表达有关;高分化组与低分化组的PTEN蛋白强阳性表达例数分别为13例、5例,高分化组PTEN蛋白表达较低分化组高(χ2=5.578,P<0.05),即肝癌分化程度与PTEN蛋白表达有关。结论肝癌组织中PTEN基因高表达,与其PTEN基因表达基本相符。Objective To research the association of Primary liver cancer (PLC) with HBsAg positive and PTEN gene ,analy-sis the relationship between PTEN protein expression and pathological type and differentiation degree .Methods The cases of blood and postoperative fresh organization with chronic HBV infection were selected from the inpatient of Binzhou medical uni-versity affiliated hospital among 2009 to 2011 .The PTEN genes were amplificated by RT-PCR ,the immunohistochemical tes-ting of antiviral sheep PTEN polyclonal antibody were chosen .Results PCR amplification of PTEN gene which located in 254 bp PTEN gene amplified bands and 380 bp GAPDH reference gene bands were got in liver cancer tissue .The PTEN-positive de-tection rate was 69.0% (20/29) in foci organization ,the relative expression was 0.713 ± 0.125 .PTEN protein expression im-munohistochemistry results showed that the PTEN-positive staining was located in the cytoplasm ,showing brown yellow granu-lar or flake .In the expression level ,the (+ ) expression of PTEN protein was 4 cases ,the (++ ) expression of PTEN protein was 13 cases ,the (+++ ) expression of PTEN protein was 5 cases .The number of strong PTEN protein expression in hepatic cellular cancer group and cholangiohepatoma group was respectively 16 and 2 ,differences between the two groups was statisti-cally significant (χ2 =4.398 ,P〈0.05) ,namely pathologic types and PTEN protein expression had correlation .The number of high PTEN protein expression in high-differentiated group and low-differentiation group was respectively 13 and 5 .Differences between the two groups was statistically significant (χ2 =5.578 , P〈0.05) ,namely differentiation degree of liver cancer had correlation with PTEN protein expression .Conclusions PTEN gene were highly expressed in foci organization ,which was con-sistent with those PTEN gene .
