摘  要:甜菊糖又称甜菊糖甙或甜菊素,其含量的测定在我国现行标准中采用容量法和高效液相色谱法。早期甜菊糖总甙中以甜菊甙为主成分,容量法测定含量也就建立在这一基础之上,但是近年来甜菊叶品种不断改良,其总甙主成分发生改变,瑞鲍迪甙A成为主成分,因此在实际检测中使用容量法所测结果与真实值便出现了偏差。试验通过高纯度甜菊甙和瑞鲍迪甙A单一成分按照不同比例混合,经容量法和高效液相色谱法分别测定其含量,结果表明随着瑞鲍迪甙A所占比例的增加,测定结果与真实值的偏差越大。经分析这可能与瑞鲍迪甙A分子质量较甜菊甙大有关,同等质量的瑞鲍迪甙A物质的量比甜菊甙要小,容量法滴定时消耗的碱液量就小,测定结果就会偏低。而高效液相色谱法就不会受此因素的影响,测定结果也更接近真实值。Stevia sugar was also called Steviosides or Steviosin. The volumetric method and HPLC were the current standard to determine its content in our country. Stevioside (STV) was considered to be the principal component of total Steviosides early times, and the volumetric method was set up based on this theory. But the components of total Steviosides had already changed along with the constantly improvement of stevia. And rebaudioside A (Reb-A) has been the principal component in recent time. So there was a deviation between analysis results and true value with volumetric method. In this experiment, the STV and Reb-A of high purity were mixed with different ratio, then the content was determinated with volumetric method and HPLC. The result showed that the deviation became larger with the increase of Reb-A's ratio. This may be related to the higher molecular weight of Reb-A than STV. The mole number of Reb-A was less than STV with the same quality. The consumption of alkali liquor decreased. And the analysis results were lower in the end. While the HPLC method cannot be affected by this factor, so determination results were closer to the true value with HPLC.
