mTOR/P70S6K 信号通路在胃癌中的表达及临床意义

摘　　要：目的：研究 mTOR/ P70S6K 信号通路在胃癌中的表达，探讨其在胃癌发生发展中的作用及意义。方法用逆转录聚合酶链反应（RT-PCR）技术，免疫印迹技术检测26例胃癌患者癌组织、癌旁胃组织及16例非胃癌胃组织中 mTOR 及 P70S6K 的表达情况；并分析 mTOR 及 P70S6K mRNA 的表达与相关临床参数的关系。结果 mTOR 及 P70S6K 在胃癌组织中的表达水平显著高于癌旁胃组织和正常胃组织中的表达水平。mTOR mRNA和 P70S6K mRNA 在胃癌组织中的表达呈正相关，且两者在癌旁胃组织及正常胃组织中的表达亦呈正相关性。mTOR 及 P70S6K mRNA 在胃癌组织中的表达水平与病理分期、有无淋巴结转移等明显相关，而与肿瘤直径、性别等无明显关系。结论 mTOR/ P70S6K 信号通路在胃癌组织中特异性激活。mTOR/ P70S6K 信号通路可能在胃癌的发生、发展中起重要作用。Objective To study the expression of mTOR/ P70S6K signaling pathway in human gastric carcinoma,and to investigate the effect and clinical significance of mTOR/ P70S6K signaling path-way in the occurrence and development of gastric carcinoma. Methods RT-PCR and western-blot were employed to determine the expression of mTOR/ P70S6K signaling pathway in cancerous and paracancerous tissuess of 26 cases and normal healthy tissues of 16 cases. The relationship between expression of the genes mRNA in gastric carcinom tissues and related parameters was analyzed. Results The expression level of mTOR/ P70S6K signaling pathway was markedly higher in the gastric carcinoma tissues than that in the paracancerous tissues or normal stomach tissues. There was a positive correlation between mTOR mRNA and P70S6K mRNA expression in gastric carcinom tissues. There was also a positive correlation between mTOR mRNA and P70S6K mRNA expressions in paracancerous tissues and the normal stomach tissues. The expression of mTOR/ P70S6K signaling pathway in the gastric carcinom tissues was significantly corre-lated with the pathological grading,lymph node metastasis,but not with the diameter and gender. Conclu-sions The mTOR/ P70S6K signaling pathway is specifically activated in gastric carcinom tissues. The mTOR/ P70S6K signaling pathway might play an important role in the pathogenesis of gastric carcinoma.
