摘 要:目的:建立高效液相色谱-蒸发光散射检测器(HPLC-ELSD)测定保健食品中低聚木糖(以木糖计)的方法。方法:样品经醇沉去除杂质,后用硫酸在沸水浴中将低聚木糖水解为木糖,以乙腈-水(80∶20,v/v)为流动相,糖分析柱(4.6 mm×250 mm,5μm)为固定相,通过蒸发光散射检测器的检测,同一样品水解前后木糖含量的差值即为样品中低聚木糖的含量。结果:低聚木糖在0.6796~5.097μg范围内呈良好的线性关系,r=1,平均加样回收率为101.66%,RSD为1.39%(n=9)。结论:该方法快速、简便、灵敏、重现性好,可作为测定保健食品中低聚木糖的方法。Objective: To develop an HPLC-ELSD method for the determination of xylooligosaccharides in health foods. Method: The sample was removed impurity by alcohol precipitation method and the xylo-oligosaccharides was hydrolyzed into xylogyranose with sulfuric acid solution in boiling water bath, and then sapareted on a carbohydrate column with a mixture of acetonitrile-water ( 80 : 20, v/v ) as mobile phase. Detected by ELSD detector, the xylo-oligosaccharides content was the D-value of xylogyranose content before and after the hydrolyzation. Result : The linear ranges of ferulic acid was 0.6796-5.097 μg. The average recovery rate for xylo-oligosaccharides was 101.66%, and RSD was 1.39% ( n=9 ). Conclusion : The developed method is rapid, simple, sensitive and replicahle, which can be used for the determination of xylo-oligosaccharid in health foods.
