摘  要:采用响应面试验优化了凝结芽孢杆菌NJ39生长和产芽孢的发酵条件,得到芽孢生成与发酵温度、pH值、接种量和发酵时间的关系方程,求解二次多项回归方程确定了发酵温度34℃、接种量13.5%(种子液菌体密度104 cfu/ml)、pH7.1和发酵时间58h为产芽孢的最优条件,芽孢数大于2×108cfu/ml。探讨了载体不同添加量的芽孢菌泥含水量随时间变化的关系,得到在载体加量为1倍、冷阱温度-45℃和真空度20Pa的条件下,冷冻干燥10h,菌粉含水量在5%以下,芽孢数量达到1010cfu/g。In the method of response surface, the fermentation condition was improved for the growth of the Bacillus Coagulans N J39 and its production of spores. An equation of the relation among spore generation, fermentation temperature, inoeu lation, pH value and fermentation time was determined. Results of the two multiple regression equation showed that the optimal condition for producing spores was 34℃ for the fermentation, inoculation of 13. 5% (the density of bacteria was 104 cfu/ml), pH 7.1 and 58 h for the fermentation. In this condition, the quantity of spores was greater than 2× 10^8 cfu/ml. It found that, with the different dosage carrier, the moisture level of bacillus coagulans mud changed over time. The number of spores was able to reach to 10^10 cfu/g in the following condition: vacuum degree 10 torr , freeze-dried 10 hours, and less than 5% double dosage of carriers, cold trap temperature of -45℃, lyophilized powder moisture level .
