摘  要:目的探讨大豆多肽对前列腺癌PC-3细胞增殖及凋亡的影响,为临床应用大豆多肽治疗前列腺癌提供实验依据。方法用不同浓度的大豆多肽(5、10、15、20μmol/L)处理前列腺癌PC-3细胞不同时间(24、48、72 h),采用MTT法检测细胞的增殖活力,倒置显微镜观察细胞的形态,流式细胞术检测细胞凋亡率及细胞周期。结果大豆多肽可抑制前列腺癌PC-3细胞的增殖,将细胞周期阻滞在G2/M期,诱导细胞凋亡,中晚期细胞凋亡趋势明显,且呈明显的剂量与时间依赖性;随着大豆多肽浓度的增加,前列腺癌PC-3细胞密度逐渐降低,边缘趋于圆滑,细胞间隙逐渐增大,局部可见部分已固缩的细胞及死亡的细胞碎片。结论大豆多肽可抑制前列腺癌PC-3细胞增殖,诱导细胞凋亡,在临床治疗前列腺癌方面具有广阔的应用前景。Objective To investigate the effect of soybean peptide on proliferation and apoptosis of prostate cancer PC-3cells and provide an experimental basis for treatment of prostate cancer with soybean peptide in clinic.Methods PC-3cells were treated with soybean peptide at various concentrations(5,10,15 and 20 μmol / L)for various hours(24,48 and 72 h),then determined for proliferative activity by MTT assay,observed for morphology under inverted microscope,and analyzed for apoptosis rate and cell cycle by flow cytometry.Results Soybean peptide inhibited the proliferation of PC-3 cells,arrested the cell cycle at G2/ M phase,and induced cell apoptosis.The PC-3 cells at middle and late stages showed a significantly dose-and time-dependent apoptotic tendency.The density of PC-3 cells decreased gradually with the increasing soybean peptide concentration,while the cell edge became smooth,and the gap between cells was enlarged gradually.Partial pyknotic or dead cell debris were observed.Conclusion Soybean peptide inhibited the proliferation and induced the apoptosis of PC-3 cells significantly,which was of a wide prospect in treatment of prostate cancer in clinic.
