摘 要:目的分析宁波地区妇女宫颈癌组织中HPV 16型E6基因的变异情况。方法收集宫颈组织石蜡标本80份,按诊断时的年龄分组,其中年龄≤35岁为A组(37例);年龄>35岁为B组(43例)。从宫颈癌组织石蜡样本中提取HPV病毒基因组DNA,采用PCR技术扩增HPV16型E6DNA,并进行DNA测序。结果 HPV 16型E6 DNA总阳性率为43.75%,其中A组阳性率为56.76%,B组阳性率为32.56%,两组差异有统计学意义(<0.05);与德国标准株相比,本次研究共发现9个突变位点,其中178位点突变频率最高,A与B组178位点突变率差异无统计学意义(=0.70)。结论 HPV16型引起的宫颈癌有年轻化趋势,T178G突变可能是HPV 16型持续性感染的重要因素,在宫颈癌的防治中应当引起重视。Objective ToinvestigatetheHPV16 E6 genemutationincervicalcarcinomatissueinNingbodistrict.Methods Eighty cases were divided into two groups according to the patients' age at diagnosis: group A(37 cases,at age≤35)and group B(43 cases, at age〉 35). The HPV viral genomic DNA was extracted from the paraffin section of cervical cancer tissue. HPV 16 E6 DNA was amplified by PCR technique sequenced. Results The positive rate of HPV 16 E6 gene was 43.75%, respectively 56.76% in group A and 32.56% in group B. The difference was statistically significant( P〈0.05). Compared with the DNA sequence of German standard strain, the results showed that nine mutation types were detected, of which the mutation rate was highest in site 178, but the difference was not statistically significant between two groups(P =0.70). Conclusions The prevalence of the cervical carcinoma caused by HPV 16 tends to be younger. T178G mutation may be an important factor of HPV 16 persistent infection. More attention should be paid to this mutation type for prevention and treatment of cervical cancer.
