摘 要:目的检测EGFR和C-erB-2在肺腺癌组织中的表达从而探讨EGFR和C-erB-2在肺腺癌中与淋巴结转移与否的关系。方法选取67份肺癌组织标本,其中有淋巴结转移38例,无转移29例,30份肺良性病变组织标本。标本常规石蜡包埋免疫组化学染色,检测EGFR、C-erB-2、E-cadherin蛋白表达,qRT-PCR分别检测EGFR和C-erB-2的mRNA表达。分析EGFR、C-erB-2表达与肺腺癌淋巴结转移的关系。结果肺腺癌组织中EGFR、C-erB-2、E-cadherin蛋白及mRNA表达阳性率均明显高于肺良性组织,差异有统计学意义(p<0.05)。EGFR、C-erB-2、E-cadherin蛋白及mRNA表达在淋巴结转移组织中的表达均明显高于无淋巴结转移组织。EGFR、C-erB-2两者表达呈正相关,差异有统计学意义(p<0.01),而EGFR、C-erB-2蛋白表达与E-cadherin蛋白表达呈负相关,差异有统计学意义(p<0.01)。结论 11.EGFR、C-erB-2蛋白在肺腺癌组织中呈高表达,两者表达呈正相关性,且二者表达与E-cadherin表达呈负相关性。2两者可能在肺腺癌淋巴转移过程中有着密切的关系,检测EGFR、C-erB-2水平有有助于肺腺癌的病情判断。Objective To investigate the expressions of EGFR and C-erB-2 in lung adenocarcinoma and to explore the role of EGFR and C-erB-2 in the metastasis and invasion of lung adenocarcinoma.Methods 67 cases of lung adenocarcinoma were analyzed and 30 cases of normal lung samples were studied as control group. SP method was used to detect the expression of EGFR、C-erB-2 and E-cadherin in all the cases, and qRT-PCR assay was employed to detect the expression of EGFR and C-erB-2.Results The expression of EGFR and C-erB-2 in lung adenocarcinoma was significantly higher than that in normal lung samples (p 〈 0.05). The positive rate of EGFR、C-erB-2 in non-metastatic group was lower than that in metastatic group (p〈0.05).The expression of EGFR in was reversely related to C-erB-2 (p〈0.01). The expression of E-cadherin in was reversely correlated with EGFR and C-erB-2(r=-0.510, p 〈 0.01; r=-0.301, p〈0.01).Conclusion ①The over-expression of EGFR and C-erB-2 and the positive correlation between them were shown in lung adenocarcinoma, and the expression of E-cadherin in was reversely correlated with EGFR and C-erB-2. ②The over-expression of EGFR and C-erB-2 detected in lung adenocarcinoma is possibly related to metastasis and invasion of carcinoma, and they may be useful markers for determine the biological aggressiveness of lung Aden carcinoma.
