胃癌组织中AEG-1、Cyclin D1的表达及其意义

摘　　要：目的观察胃癌组织中星形细胞上调基因-1(astrocyte elevated gene-1,AEG-1)、Cyclin D1的表达,分析二者表达与胃癌侵袭、转移的关系。方法应用免疫组化EnVision法检测110例胃癌组织中AEG-1和Cyclin D1的表达,并复习相关文献。结果AEG-1在胃癌组织中的阳性率为69.1%,AEG-1表达与癌组织Lauren分型、淋巴结转移、浸润深度和pTNM分期密切相关(P<0.05),与患者性别、年龄、发生部位和肿瘤大小无关(P>0.05);Cyclin D1在胃癌组织中的阳性率为57.3%,Cyclin D1表达与癌组织浸润深度、淋巴结转移和pTNM分期有关(P<0.05),与患者性别、年龄、发生部位、肿瘤大小和Lauren分型无关(P>0.05)。相关性分析显示,胃癌组织中AEG-1与Cyclin D1的表达呈正相关(P<0.05)。结论 AEG-1可能通过上调Cyclin D1的表达而促进胃癌细胞的侵袭、转移。AEG-1可能作为胃癌治疗的一种潜在分子靶点。Purpose To study the expression of AEG-1 and Cyclin D1 in gastric adenocarcinoma, and to explore the relationship be- tween AEG-1, Cyclin D1 expression and invasion, metastasis of gastric carcinoma. Methods Expression of AEG-1 and Cyclin D1 in 110 cases of gastric adenocarcinoma was detected by EnVision immunohistochemical technique. Results The positive rate of AEG-1 was 69. 1% in gastric carcinoma. Expression of AEG-1 was related to Lauren type, lymph node metastasis, the depth of invasion, and pTNM stage （P 〈 0. 05 ）, but unrelated to sex, age, location, and size （P 〉 0. 05 ）. The positive rate of Cyclin D1 was 57.3 % in gas- tric carcinoma. Cyclin D1 expression in gastric carcinoma was correlated with the depth of invasion, lymph node metastasis, and pTNM stage （P 〈 0. 05） , however, there was no relationship between Cyclin D1 expression and sex, age, location, tumor size, and Lauren type （ P 〉 0. 05 ）. Correlation analysis showed there was a positive correlation between AEG-1 expression and Cyclin D1 expression （ P 〈 0.05）. Conclusions AEG-1 may contribute to the invasion and metastasis of gastric carcinoma by upregulating Cyclin D1 expres- sion. AEG-1 may be a potential molecular target for the treatment of gastric adenocarcinoma.
