摘 要:为了获得单一的细菌素Enterocin E9成分,使其发挥最大抑菌活性,建立在线SPE/HPLC分离Enterocin E9的方法。通过细菌培养,上清液收集、浓缩,获得细菌素粗提样品,采用Sepbox 2D-250全自动二维液相分离系统,确认流动相组成,流速和洗脱时间等因素。提取的单一成分其抑菌效果是浓缩样品的2.4倍。该方法省时、省力、精密度高,为细菌素结构与功能的研究提供一条有效途径和方法。The aim of this work was to obtain the purified Enterocin E9 with the highest antibacterial activity.A new method based on on-line solid-phase extraction (SPE) coupling to high-performance liquid chromatography (HPLC) for the determination of Enterocin E9,has been established.The extracts were finished by bacterial culture,supernatant collection and concentration respectively.The validation parameters such as the composition of mobile phase,flow rate and elution time et al,were detcted by Sepbox 2D-250 automatic two-dimensional liquid separation system.The optimization and validation of a SPE/HPLC for the separation and purification of Enterocin E9 was identified.The activity of homogeneity was as much as 2.4 times higher than that of those concentration samples.In conclusion,it indicated This new method is rapid,simple,and sensitive.It could be an efficient way for further studying the bacteriocin structure and function.
