摘 要:目的:建立大孔树脂同时分离纯化枇杷叶中熊果酸和科罗索酸的工艺条件。方法:实验采用乙醇浸提法提取枇杷叶中熊果酸和科罗索酸,经活性炭脱色,采用大孔吸附树脂层析同时分离纯化枇杷叶中熊果酸和科罗索酸。结果表明:提取液经活性炭脱色后色素去除率达91.1%,熊果酸回收率为90%,科罗索酸回收率为91.5%。采用HZ816树脂进行柱分离,最适工艺为:上柱流速为1BV·h-1,上样液熊果酸浓度为2mg·m L-1;洗脱流速为1BV·h-1,洗脱剂的乙醇体积分数顺序为38%-55%-95%,分离后得到熊果酸和科罗索酸纯度分别为85.6%和52%。结论:建立的工艺条件可同时分离枇杷叶中熊果酸和科罗索酸,且收率较高,适宜工业化生产。Objective To optimize separation and purification technology of ursolic acid and corosolic acid from Ioquat leaf by macroporous adsorption resin. Methods=The present experiment extracted ursolic acid and corosolic acid from Ioquat leaf by ethanol, Decolorizing with active carbon,and purified ursolic acid and corosolic acid by macroporous adsorption resin chromatography separation,and then evaluated. Results=Extract the active carbon decoloring pigment removal rate 91.1% ,90% ursolic acid and 91.5% corosolic acid reclaimed. The optimum technological conditions of separation by HZ816 macroporous resin were 1BV~ h 1 loading speed, 2mg~ mL-~ ursolic acid loading concerntration, 1BV. h-1 eluting speed and the volume fraction of ethanol elution order 38%-55%-95%,after the separation obtained ursolic acid and corosolic acid purity was 85.6% and 52%. Conclusion=These established process conditions could be adopted to separate ursolic acid and corosolic acid at the same time with high yield ,which were suitable for industrial production.
