摘  要:胶原蛋白因具有独特的氨基酸组成和结构,被广泛地应用于医疗、食品及美容等领域。选取鹿皮为原料,通过酸水解和酶水解结合法提取胶原蛋白,其提取率可达32.38%,进一步分离纯化,通过测定原料中羟脯氨酸的含量换算后得出其所含胶原蛋白含量为9.912%。通过SDS-PAGE测定胶原蛋白的分子量为30万,并对其自由基清除能力进行了研究,在测定范围内,随着胶原蛋白浓度的增加,其自由基清除能力均增大,高浓度时其清除能力较强。Collagen is widely used in medical, food and cosmetic fields because of its unique amino acid composition and structure. Deer skin was selected as raw materials to extract collagen through combination with acid hydrolysis and enzymatic hydrolysis,such method could achieve 32?38% ex?traction rate of collagen. Then it was separated and purified. Content of hydroxyproline in raw materi?als was determined and content of its collagen was obtained through conversion, which was 9?912%. Molecular weight of collagen was measured by SDS-PAGE,which was about 300 thousand, and its free radical scavenging capacity was studied. Within the range of measurement,the free radical scav?enging capacity increased as collagen concentrations increased. Collagen had strongly scavenging ca?pacity at high concentration.
