摘　　要：目的探讨Orexin A对抑郁症模型大鼠的学习记忆能力的影响。方法实验大鼠随机分为空白组、空白对照组、抑郁对照组、抑郁治疗组。除空白组,其余3组实验前施以杏仁体埋管手术。抑郁对照组、抑郁治疗组采用孤养加慢性不可预见性的温和应激(Chronic Unpredictable Mild Stress,CUMS)诱导抑郁症大鼠模型,分别采用旷场实验、糖水偏爱百分比及体重及摄食量测定对抑郁大鼠模型评分。空白组和空白对照组不注射任何试剂,抑郁治疗组给Orexin A,抑郁对照组给等量生理盐水,采用平衡木和Morris水迷宫测试记录大鼠学习记忆成绩。结果空白对照组的水平得分、垂直得分、糖水偏爱百分比、体重和摄食量与空白组相比,差异均无统计学意义(P>0.05);而抑郁对照组和抑郁治疗组的水平得分、垂直得分、糖水偏爱百分比、体重和摄食量与空白对照组相比,差异均有统计学意义(P<0.05);抑郁治疗组的平衡木和Morris水迷宫测试结果均短于抑郁对照组,差异有统计学意义(P<0.05)。结论采用孤养加CUMS可建造大鼠抑郁症模型,Orexin A能够改善抑郁症模型大鼠的学习记忆能力。Objective The aim of this study was to explore the effect of Orexin A on learning ability and memory of depression model rat.Methods The experimental rats were randomly divided into blank group,blank control group,depression control group,and depression treatment group.Except for the blank group,the pipes were buried under the amygdala of the rats in three other groups before experiments.For the depression control group and the depression treatment group,the depression model rats were induced by solitary keep combined with chronic unpredictable mild stress（CUMS）.The depression model rats were evaluated by open-field test,sucrose water percentage preference,body weight and food intake.The blank group and the blank control group were not injected with any reagent.The depression treatment group was administered with Orexin A,and the depression control group was treated with the same volume of saline.The learning ability and memory of rats was assessed by the balance beam and Morris water maze test.Results Compared with the blank group,the blank control group showed no statistical significant difference in the level score,vertical score,sucrose water percentage preference,body weight and food intake（P〉0.05）.Compared with the blank control group,the depression control group and depression treatment group displayed statistical significant difference in the level score,vertical score,sucrose water percentage preference,body weight and food intake（P〈0.05）.The balance beam and Morris water maze test results for depression treatment group were shorter than the results for the depression control group,and this difference was statistical significance（P〈0.05）.Conclusion The depression model of rats can be built by the solitary keep combined with CUMS.Orexin A can improve the learning ability and memory of depression model rat.
