摘 要:以意大利苍耳叶为原料,研究其总酚的提取条件,同时对意大利苍耳叶总酚提取物的抗氧化活性进行评价。选用乙醇-水混合物为溶剂,采用超声辅助提取法处理意大利苍耳叶。将中心组合实验设计原理和响应面分析方法相结合,模拟得到二元多次相回归方程的预测模型和可信度,并通过岭脊分析方法得到最佳提取条件。使用福林酚试剂对意大利苍耳叶的总酚含量进行测定。分别采用铁离子还原法、DPPH自由基清除法测定意大利苍耳叶提取物的抗氧化能力。结果表明:超声波辅助提取意大利苍耳叶总酚的最佳工艺条件为乙醇体积分数54%、超声时间31min、液固比195mL·g-1;意大利苍耳叶总酚提取率为(3.12±0.27)%。意大利苍耳叶总酚提取液对铁离子还原的IC50为(774.5±7.26)mg·L-1,对DPPH自由基的半数抑制率IC50为(250.9±5.48)mg·L-1。由响应面法优化得到的意大利苍耳叶总酚的提取工艺方便可行,得到的意大利苍耳叶总酚具有一定抗氧化活性。Using the leaves of X.italicum Moretti as raw material,the extraction conditions of total phenols and antioxidant ac-tivity of the total phenolic extract was investiged.Using ethanol-water mixture as solvent,the leaves of X.italicum Moretti were processed by ultrasonic assisted extraction.The center combination experiment design principle and the analysis of the response surface method were combined to get the forecast model of binary regression equation of the multiple phase and credibility,and the best extraction conditions were obtained through ridge analysis method.The total phenolic content was determined by the Fo-lin-phenol reagent.Antioxidant capacity was measured using iron reduction method and DPPH radical scavenging method respec-tively.The results show that the optimum conditions of ultrasonic assisted extraction are ethanol concentration 54%,extraction time 31 min,liquid-solid ratio 195 mL·g-1 ,and yield of total phenols is (3.12±0.27)%.The leaves of X.italicum Moretti on the reduction of iron ions IC50 was (774.5±7.26)mg·L-1 and DPPH IC50 is (250.9±5.48)mg·L-1 .The response surface method to optimize the extraction process of total phenols are convenient and feasible,and the X.italicum Moretti leaves total phenol has certain antioxidant activity.
