摘  要:目的建立基于UPLC-MS/MS与固相萃取(SPE)相结合的大鼠血浆中蒜氨酸的分析方法。方法用强阳离子基质的SPE柱提取大鼠血浆中的蒜氨酸和降低基质效应,UPLC-MS/MS法进行测定。结果对SPE方法中淋洗液和洗脱液进行优化,选择水为淋洗液,甲醇-氨水(95∶5)为洗脱液,回收率>68%;蒜氨酸的标准曲线相关系数r2>0.99;蒜氨酸测定的日内精密度和日间精密度良好,相对标准偏差(RSD%)分别低于2.1和8.2。结论本方法可有效降低复杂生物样品中的基质效应,测定方法准确、可靠,具有很好的应用前景。Objective To establish an analysis method of alliin in rat plasma based on UPLC/MS/MS in combination with solid phase extraction (SPE).Methods SPE column (strong)cationic matrix was used to extract alliin in rat plasma and to reduce the matrix effect,with UPLC/MS/MS method for determina-tion.Results Optimize the eluent in SPE method and choose water as eluent,methanol-ammonia (95:5) as the eluent,with the recovery rate above 68%,the correlation coefficient of Alliin standard curve r2〉0.99,good inter-and intra-day precision for the alliin determination,and the relative standard deviation (RSD)less than 2.07 and 2.07 respectively.Conclusion This method is effective in reducing the matrix effect of complex biological samples,with accuracy,reliability and good application prospects.
