摘 要:为获得含量较高的五味子多糖,采用响应面优化试验法、正交试验法分别对五味子多糖的提取、脱色及脱蛋白工艺进行优化。结果表明:五味子多糖的最佳提取工艺条件为药材粉碎过筛60目,料液比1∶30(g/mL),超声功率165.31W,萃取38.75min;活性炭脱色最佳工艺为调节pH 5.0,脱色温度80℃,活性炭用量1.5%,搅拌30min;脱蛋白的最佳工艺条件为加入Sevag试剂(氯仿∶正丁醇=4∶1),每次振荡20min,样液与试剂比为5∶1,重复处理5次。在此优化条件下,五味子多糖提取率达10.52%,纯度由41.76%增加到79.53%。The extraction and purification technology conditions of polysaccharides fromS.chinensis was optimized to get the highest content of polysaccharides.Response surface methodology and orthogonal test were employed to optimize the extraction,decoloration and deproteinization conditions,respectively.Results:The optimum extraction conditions with ultrasound-assisted method were particle size of sample sieved with 60 mesh,ultrasound power of 165.31 W,solid to liquid ratio of 1∶30and extraction time of38.75min;The optimum decoloration conditions were pH of 5.0,temperature of 80℃,amount of active carbon of 1.5%,shaking time of 30min;and the optimum deproteinization conditions were Sevag reagent(chloroform/normal butanol=4∶1),shaking time of 20 min,sample liquid to Sevag regent ratio of 1∶5and deproteinization times of 5times.Under these optimized conditions,the extraction efficiency of polysaccharides reached 10.52%,and the purity of polysaccharides increased from 41.67%to 79.53%.
