摘 要:目的分析人乳腺组织中P-JNK和beclin 1的表达变化,探讨二者在乳腺癌鉴别诊断及治疗中的临床作用。方法应用免疫组化SP法检测20例乳腺正常上皮、30例乳腺导管原位癌和70例乳腺浸润性导管癌中P-JNK和beclin 1蛋白的阳性率,Western blot检测乳腺浸润性导管癌及正常乳腺组织中P-JNK和beclin 1蛋白的表达量。结果 1在正常乳腺组织、乳腺导管原位癌和乳腺浸润性导管癌中P-JNK阳性率分别为80%、43.3%和18.6%,beclin 1阳性率为85%、46.7%和24.1%。2乳腺癌中二者阳性率与组织学分级相关(P<0.05),与患者临床分期、淋巴结转移、ER和PR阳性率无关(P均>0.05)。370例乳腺癌组织中,P-JNK和beclin 1表达呈正相关性(P<0.01)。4Western blot结果表明,P-JNK和beclin 1蛋白在乳腺癌组织中蛋白表达量低于二者在乳腺正常组织中的蛋白表达量(P<0.05)。结论在乳腺癌中P-JNK和beclin 1均低表达,P-JNK和beclin 1具有正相关性,二者可能在乳腺癌发生、发展过程中受到抑制,因此推断联合检测P-JNK和beclin对于乳腺良、恶性病变的鉴别以及对乳腺癌的治疗、评价预后有重要参考价值。Objective To investigate the expression of P-JNK and beclin 1 in breast tissues and their relation in the breast carcinoma. Methods The expression of P-JNK and beclin 1 was examined in 120 cases of breast paraffin- embbeded specimens using immunohistochemical SP method and the relationship between the P-JNK and Beclinl expression and the clinical pathologic features. The breast cancer and pairing breast normal tissue were subjected to Western blot analysis for P-JNK and beclinl protein. Results P-JNK and beclinl expression significantly decreased during breast carcinogenesis and was correlated with histologic grade but not with stage, lymphatic metastasis, and status of ER and PgR (P 〉 O. 05). The immunohistochemical results showed that there was an decreasing tendency in positive expression rate of P-JNK and beclin 1 in NBT, DCIS, and IDC, respectively, that was, 80. 0%, 43.3% , 18.6% and 85.0% , 46. 7% , 24. 1%. Spearman~ correlation revealed that P-JNK expression was significantly correlated with beclin 1 expression in the 70 breast cancer specimens ( rs = O. 646, P 〈 0. 001 ). Western blot analysis showed that P-JNK and beclin 1 protein expression was significantly higher in breast normal tissues compared with breast tumor tissues. Conclusion The results strongly suggest P-JNK and beclinl are both decreased expression in human breast cancer. We conclude that in breast cancer cells increased expression of P-JNK would make the expression of beclin 1 increased.
