健脑安神胶囊对失眠大鼠EEG、EOG、EMG的睡眠时相及安全性的相关实验研究

摘　　要：目的:观察健脑安神胶囊对失眠大鼠睡眠时相及肝、肾功能的影响。方法:将80只Wister大鼠,随机分为假手术组、模型组、中药模型治疗组、西药模型治疗组各20只,采用对氯苯丙氨酸(PCPA)腹腔注射复制失眠大鼠模型,各组灌胃治疗3周,治疗前后监测各组大鼠脑电图(EEG)、眼动电图(EOG)、肌电图(EMG)及肝、肾功能的变化。结果:中药模型治疗组与模型组相比觉醒期(Waking)明显缩短,慢波睡眠期(SWS)、快动眼睡眠(REMS)、总睡眠期(TST)明显回升(P<0.01)。比较4组治疗前后尿素氮、肌酐、谷丙转氨酶、谷草转氨酶,无统计学意义(P>0.05)。结论:健脑安神胶囊治疗失眠的疗效确切,其治疗失眠的作用机制与减少失眠大鼠的觉醒时间,延长SWS、REMS、TST期有关。且该药物对大鼠肝肾功能无明显损害,具有良好的药物安全性。Objective： To observe the effect of Jiannao Anshen Capsule on the sleep phase and the functions of insomnia rats＇ livers and kidneys. Methods： 80 Wister rats were randomly divided into sham operation group,model group,Chinese medicine treatment model group,western medicine treatment model group and each model had 20 rats. Insomnia rat models were made by injecting the p-chlorophenylalanine（ PCPA） to intraperitoneal region. All rats were given intragastric administration for three weeks. Before and after the treatments,electroencephalogram（ EEG）,electrooculogram（ EOG） and electromyogram（ EMG） were tested and the changes of functions of livers and kidneys were monitored. Results： Chinese medicine treatment group got significantly shorter time in waking and longer time in slow wave sleep（ SWS）,rapid eye movement（ REM） and total sleep time（ TST） than model group（ P〈0. 01）. There were no significantly differences among the fore groups in BUN,CR,ALT,AST by paired sample t test before and after the treatments. Conclusion： Jiannao Anshen Capsule has an exactly effect in the treatment of insomnia. The functions of the treatment of insomnia are related with the decrease of insomnia rats awakening time and prolonging SWS,REMS,TST phase. The drug has no obvious damage on the functions of rats＇ livers and kidneys and has good security.
