摘 要:目的:探讨结直肠癌( CRC)组织中血管紧张素转换酶( ACE)基因多态性。方法对30例符合要求的CRC患者,应用PCR检测手术切除的肿瘤组织和癌旁正常组织中ACE基因I/D多态性分布。结果CRC组织中II型、ID型、DD型3种基因型的频率分别为16.67%、43.33%、40.00%,癌旁正常组织分别为30.00%、53.33%、16.67%(χ2=3.857,P〈0.05)。CRC组织中I等位基因和D等位基因分布频率分别为38.33%及61.67%,癌旁正常组织分别为56.67%和43.33%,两者比较的OR值为1.423(1.001~2.002)(P〈0.05)。结论 CRC组织中ACE基因DD亚型高表达及D等位基因分布频率异常与CRC关系密切,D等位基因高表达可能是CRC易患因素之一。Objective To investigate the polymorphism of angiotensinⅠ-converting enzyme( ACE)gene in the patients with colorectal cancer( CRC). Methods Thirty patients with CRC who received surgery treatment were enrolled in the present study. PCR method was used to assay insertion/deletion genotypes and I/D alleles of ACE gene in the tumor tissue and para-tumor normal tissue. Results The frequencies of genotypes of II,ID and DD were 16. 67%,43. 33%,40. 00% of the CRC tissues,those were 30. 00%,53. 33%,16. 67% of the paraneoplastic normal tissues(χ2 =3. 857,P〈0. 05). The frequencies of I and D alleles were 38. 33% and 61. 67% respectively, and were 56. 67% and 43. 33% respectively,the OR value was 1. 423(1. 001-2. 002)(P〈0. 05). Conclusion The high expression of DD genotype and abnormal distribution of D allele of ACE gene are related to CRC,and high D allele ratio can be the carcinomatous change factor of CRC.
