Detection and Quantification of Melatonin from Aloe vera, Medlar Leaves and Mulberry Leaves by HPLC-UV

摘  要:建立提取和检测芦荟、枸杞叶和桑叶的中褪黑素的方法。以甲醇为提取剂,采用超声波法提取褪黑素。色谱柱为Agilent Eclipse XDB-C18(250 mm×4.6 mm,5μm),流动相为无水甲醇-水(50∶50,V/V),流速为1.0 m L/min,检测波长为222 nm,柱温为室温。结果表明,在所建立色谱条件下,褪黑素质量浓度在2.136×10^1-2.136×104ng/m L范围内,浓度和峰面积的线性关系良好(R2=0.999 5),最低检出限为0.2 ng。经测定芦荟中褪黑素含量为46.60ng/g,回收率为100.5%,RSD为1.2%;枸杞叶中褪黑素含量为1.235×10^4 ng/g,回收率为103.9%,RSD为1.2%;桑叶中褪黑素含量为5.452×10^4 ng/g,回收率为103.6%,RSD为1.9%。所建立HPLC-UV法简单易行,检测速度快,精密度、稳定性和准确度都令人满意。Establish the method on extracting and determining melatonin from Aloe vera, medlar leaves and mulberry leaves. Methanol as extraction agent, melatonin was extracted by ultrasonic method and then analyzed by HPLC-UV technique. Chromatographic column was Eclipse XDB-C18 (250 mm×4.6 mm, 5 μm); mobile phase was anhydrous methanol-water (50 : 50, V/V); flow rate was 1.0 mL/min; detection wavelength was 222 nm; column temperature was room temperature. Results showed that under the chromatographic condition, linear relationship between peak and concentration over the range of 2.136× 10^1-2.136×10^4 ng/mL (R2=0.999 5) was good, and the detection limit was 0.2 ng. The melatonin content of Aloe vera was 46.60 ng/g, and average recovery was 100.5% with a RSD of 1.2%. Melatonin form Medlar leaves was 1.235×10^4 ng/g, and average recovery was 103.9% with a RSD of 1.2%. Melatonin form Mulberry leaves was 5.452× 10a rig/g, and average recovery was 103.6% with a RSD of 1.9%. This HPLC-UV method was simple, rapid, precision, stable, with satisfactory accurate.
