红花黄色素、阿魏酸对痛经模型大鼠催产素受体及加压素受体mRNA表达的影响  被引量:8

Influence of safflower yellow and ferulic acid on the oxytocin receptor and vasopressin receptor mRNA expressions in the uterus of dysmenorrhea model rats

摘  要:目的探讨红花黄色素、阿魏酸在抗痛经作用中的相关作用机制。方法 70只雌性SD大鼠按体重随机分为7组:正常组、模型组、田七痛经胶囊组(田七组)、红花黄色素高剂量组(红高组)、红花黄色素低剂量组(红低组)、阿魏酸高剂量组(阿高组)及阿魏酸低剂量组(阿低组),每组10只。除正常组外,大鼠股部皮下注射己烯雌酚复制大鼠痛经模型,田七组在皮下注射己烯雌酚开始时给予田七胶囊2 g·kg-1,红高组、红低组于注射第7天分别给予红花黄色素0.02 g·kg-1·d-1、0.01 g·kg-1·d-1,阿高组、阿低组于注射第7天分别给予阿魏酸0.12 g·kg-1·d-1、0.06 g·kg-1·d-1。共注射11 d。采用酶联免疫法(ELISA)检测各组大鼠子宫组织前列腺素E2(PGE2)、前列腺素F2a(PGF2a)、血浆血栓素B2(TXB2)、6酮前列腺素F1a(6-keto-PGF1a),采用逆转录-聚合酶链反应(RT-PCR)方法检测子宫组织催产素受体(OTR)mRNA及加压素受体(VPR)mRNA的表达情况。结果与模型组比较,各药物处理组扭体反应均有所改善,但无统计学差异(P均>0.05)。与模型组比较,田七组、阿高组、阿低组PGE2含量均有不同程度升高(P<0.01或P<0.05);而田七组、阿高组、阿低组、红高组PGF2a含量及PGF2a/PGE2的比值均有不同程度降低(P<0.01或P<0.05);各药物处理组TXB2及TXB2/6-keto-PGF1a的比值均显著降低(P均<0.01)。与正常组比较,模型组大鼠子宫OTRmRNA和VPRmRNA相对表达量显著增高(P均<0.01);与模型组比较,田七组OTRmRNA、VPRmRNA相对表达量均下降(P均<0.05),阿高组OTRmRNA相对表达量下降(P<0.05);其他组组间比较无统计学差异(P均>0.05)。结论阿魏酸抗痛经作用可能与调节前列腺素系统及调节痛经大鼠子宫OTR表达有关,红花黄色素可调节前列腺素系统但对OTR及VPR表达无明显影响。Objective To investigate the related mechanism of safflower yellow and ferulic acid on anti-dysmenorrhea.Methods Seventy female rats were randomly divided into seven groups( n = 10 each) : normal group,model group,tianqi group,safflower yellow high dose and low dose groups,ferulic acid high dose and low dose groups. The dysmenorrhea rat model was established by share hypodermic injection of diethylstilbestrol in all rats but normal group. Tianqi capsule( 2 g·kg^-1) was given at the beginning of hypodermic injection of diethylstilbestrol in tianqi group; 0. 02 g·kg^-1·d^-1and 0. 01 g·kg^-1·d^-1of safflower yellow were respectively given in the seventh day of hypodermic injection in safflower yellow high dose and low dose groups; 0. 12 g·kg^-1·d^-1and 0. 06 g·kg^-1·d^-1of ferulic acid were respectively given in the seventh day of hypodermic injection in ferulic acid high dose and low dose groups. The hypodermic injections were11 days in all. Enzyme linked immunosorbent assay( ELISA) was used to detect the levels of prostaglandin E2( PGE2),prostaglandin F2a( PGF2a) in the uterus tissues and plasma thromboxane B2( TXB2),6-keto-PGF1 a. Reverse transcription-polymerase chain reaction( RT-PCR) method was used to detect the mRNA expressions of oxytocin receptor( OTR)and vasopressin receptor( VPR) in rat uterus tissues. Results Compared with model group,the body torsion reactions in drug-treated groups were all improved without statistical difference( all P〈0. 05). Compared with model group,the contents of prostaglandin E2 in tianqi group,ferulic acid high dose and low dose groups increased( P〈0. 01 or P〈0. 05),while the contents of PGF2 a and the ratios of PGF2 a / PGE2 in tianqi group,ferulic acid high dose and low dose groups,safflower yellow high dose group decreased( P〈0. 01 or P〈0. 05). The contents of TXB2 and the ratios of TXB2 /6-ketoPGF1 a decreased significantly( all P〈0. 01) in all drug-treated groups. Compared with normal group,the OTR
