摘  要:目的探讨染料木黄酮对肝癌细胞SMMC7721增殖的抑制作用及其作用机制。方法以染料木黄酮干预肝癌细胞SMMC7721。MTT法及集落形成实验检测细胞增殖;蛋白质免疫印迹检测染料木黄酮对ERK及EGFR活化的影响;免疫共沉淀法检测染料木黄酮对SMMC7721细胞ERK及EGFR间相互作用的影响。结果染料木黄酮能显著抑制肝癌细胞增殖,其抗增殖活性具有浓度依赖性;染料木黄酮还可抑制ERK与EGFR的活化,并抑制ERK及EGFR间的相互作用。结论染料木黄酮可抑制肝癌细胞增殖,其机制可能与其抑制EGFR/ERK通路活化有关。Objective To investigate the anti- proliferative effects of genistein on hepatoma SMMC7721 cells and to explore its underlying mechanisms. Methods SMMC7721 cells were treated with genistein. Cell proliferation was examined by MTT and colony formation assay. Activation of EGFR and ERK was determined by immunoblotting. Co- immunoprecipitation was employed to explore the effect of genistein on the interaction between EGFR and ERK. Results Genistein significantly suppressed the proliferation of SMMC7721 cells,which was of concentration dependence. Besides,genistein could inhibit the activation of EGFR and ERK as well as their interaction. Conclusion Genistein can inhibit the process of proliferative activity of hepatoma cells,which may be related to the suppression of EGFR / ERK pathway.
