摘 要:以一株耐高温、耐高糖的葡萄糖酸钠生产菌株Aspergillus niger SFGT601为研究对象,在前期研究工作的基础上,首先利用5L发酵罐对其发酵培养基组成进行了优化,得到了理想的培养基组成为葡萄糖320 g/L,玉米浆3.5 g/L,酵母青2.0 g/L,硫酸镁1.5g/L,然后进一步考察了pH值和溶解氧对其发酵的影响,确定了最佳的pH和溶解氧控制策略为溶解氧20%,0~8h控制pH值5.5~6.0,8~22h控制pH值5.0~5.5.在此最佳工艺条件下菌株SFGT601的葡萄糖酸钠产量提高到326.0 g/L.A high temperature and sugar tolerant sodium gluconate-producing Aspergillus niger SFGT601 was investigated in this paper.In order to further enhance its production and conversion ratio,combined with the flasks experiment,the medium components and process conditions were optimized in a 5-L fermentor.The optimal medium compositions were obtained as follows:glucose 320 g/L,corn steep liquor 3.5 g/L,yeast extract 2.0 g/L,magnesium sulfate 1.5 g/L.Then the effect of dissolved oxygen and pH value were investigated on sodium gluconate production by A.niger SFGT601,and the optimal pH and dissolved oxygen control strategy was also established.The optimal condition was as follows:dissolved oxygen 20%,pH 5.5-6.0 for 0-8 h,pH 5.0-5.5 for 8-22 h.Under the conditions of optimized fermentation medium and process parameters,the sodium gluconate production rose to 326.0 g/L.
