摘　　要：目的:研究GPC3基因在人胃癌中的表达,并结合wnt-β-catenin的表达研究其相关性,进一步研究GPC3的促瘤效应及其机制。方法:荧光定量PCR及免疫组织化学检测GPC3及wnt-β-catenin通路在人胃癌中的激活情况,胃癌细胞系SGC-7901体外研究GPC3体外促瘤效应。分别采用MTS及Annexin-PI共染法检测GPC3的促增殖抗凋亡效应。Western blot检测wnt-β-catenin信号通路的激活及下游基因的表达。结果:与癌旁正常胃组织相比,GPC3在人胃癌组织中高表达,且其表达与wnt-β-catenin信号通路呈正比。体外研究发现GPC3能够促进胃癌细胞的增殖,降低胃癌细胞的凋亡率,而其机制在于GPC3能够激活wnt-β-catenin通路及下游促癌基因。结论:GPC3基因在人胃癌组织中高表达,并通过激活wnt-β-catenin通路发挥癌基因生物学效应。Objective：To investigate the expression of GPC3 in human gastric cancer and its relation with activa-tion of wnt - β - catenin signaling,furthermore to investigate the mechanism of its tumor promotion effects. Methods：The expression of GPC3 in human gastric cancer was detected by real - time PCR and immunohistochemistry. Prolifer-ation and apoptosis of SGC7901,a human gastric cancer cell line,was investigated by MTS and PI - Annexin V co -staining assay,the activation of wnt - β - catenin signaling was accessed by Western blot. Results：Overexpression of GPC3 in human gastric cancer tissue was found compared to adjacent normal tissues,the expression of GPC3 was cor-related to the expression of β - catenin in gastric cancer. GPC3 had proliferation and survival effects in SGC7901 cells,which was depended on wnt - β - catenin activation. Conclusion：GPC3 is a oncogene with proliferation and survival effects due to wnt - β - catenin activation.
