Identification and Quantification of Methyl Jasmonate in Soybean Roots Using Headspace Solid-Phase Microextraction Coupled with GC-MS

摘  要:建立了顶空固相微萃取(HS·SPME)与气相色谱(GC)-质谱(MS)联用技术测定大豆根样品损伤后所分泌的挥发性物质茉莉酸甲酯的方法.探讨了萃取时间、萃取温度、解吸时间以及离子强度对茉莉酸甲酯萃取量的影响,并对测试参数进行优化.结果表明:优化参数是提高检测灵敏度的有效手段,茉莉酸甲酯的响应值与浓度具良好的线性关系,最小检测量是0.5μg/L(在植物中为1.5ng/g),线性范围为0.7—25μg/L(在植物中为2.1~750ng/g).该方法准确、灵敏、快速,消耗样品量少,在同类样品内源产生的茉莉酸甲酯测定中显示出独特优势,并为建立其它生物材料中茉莉酸甲酯的测定方法提供参考.A headspace solid-phase microextraction in combination with gas chromatography and mass spectrometry was used for enrichment, identification and quantification of volatile methyl jasmonate secreted by wounded soybean root. The test parameters such as extraction time, extraction temperature, desorption time and ionic strength were studied and optimized. There was a good linear relationship between the concentrations and peak area values of methyl jasmonate in the range of 0. 7 -25 μg/L (in plant is 2. 1 -750 ng/g). The detection limit of methyl jasmonate was 0. 5 μg/L (in plant is 1.5 ng/g). The method was precise, sensitive and rapid with minimum sample size. It shows unique advantage in determination of intrinsic methyl jasmonate in the similar samples, and the reference was provided for determination of the methyl jasmonic acid in other biological materials.
