摘  要:目的:探讨珊瑚状猴头菌多糖的抗氧化作用。方法:体外抗氧化活性以测定其总还原力,对DPPH自由基、O2-·、·OH的清除率,抑制体外脂质过氧化能力为指标;体内抗氧化试验:将48只昆明小鼠,雌、雄各半,随机分为对照组、低剂量组【100 mg/(kg·d)】、中剂量组【200 mg/(kg·d)】、高剂量组【400 mg/(kg·d)】4个组。灌胃30d后,采样,测定小鼠血清、心脏、肝脏、肾脏组织中总抗氧化能力、超氧化物歧化酶、谷胱甘肽过氧化物酶、过氧化氢酶活力及丙二醛含量。结果:随着珊瑚状猴头菌多糖质量浓度的增加,其抗氧化能力增强;当其质量浓度达到2 mg/m L时,还原力吸光度为0.482,对DPPH自由基、O2-·和·OH的清除率分别为80.10%,91.72%和94.33%,脂质过氧化物抑制率为68.71%;可提高心脏、肝脏、肾脏等组织及血清中T-AOC、SOD、GSH-Px和过氧化氢酶活力,降低MDA含量,在高剂量时差异达极显著水平(P<0.01)。结论:珊瑚状猴头菌多糖具有抗氧化作用。Object: To investigate the effects of polysaccharides from Hericium coralloids. Methods: Antioxidant activity of Hericium coralloids polysaccharide in vitro is determined by the total reducing power, the scavenging capacity of DPPH·, superoxide anion radical(O2^-·), hydroxyl radical(·OH) and lipid oxidation inhibiting activity; Experiment in vivo, 48 healthy mice were randomly divided into control group, low dose group(100 mg/kg), middle dose group(200mg/kg) and high dose(400 mg/kg)(female∶male=1∶1). After 30 d, the levels of MDA and the activity of T-AOC, SOD,CAT, GSH-PX in serum, heart, liver, kidney were examined. Results: Hericium coralloids polysaccharide has certain antioxidant activity. Values of the ability of antioxidant increased with increasing the concentrations of polysaccharide. At2 mg/ml, the absorbance of reducing powers was 0.482, DPPH·、 O2^-·and ·OH scavenging rate, and lipid oxidation inhibiting activity were 80.1%, 91.72%, 94.33% and 68.7% respectively. Hericium coralloids polysaccharide can increase the contents of T-AOC, SOD, GSH-Px, CAT and decrease the levels of MDA in organ and serum, and the difference were significant in high dose(P〈0.01). Conclusion: It was indicated Hericium coralloids polysaccharides had antioxidant activity.
